    Peat pots and peat pellets soaked in different concentrations of auxin were tested as an alternative to plastic pots and to improve propagation efficiency by eliminating the laborious work involved in softwood cutting propagation of ‘MKR1’ persimmon dwarfing rootstock. Cuttings in peat pots produced similar rooting percentages to those in plastic pots. The roots of cutting in pots placed on water-saturated trays were significantly longer than those placed on perforated, drained trays. Compared to intact leaves and 1/4 leaf trimming, cuttings with 1/2 of the leaf blade decreased the rooting percentage. Perlite produced 15% rooting, but when mixed with peat (1:1), it enhanced rooting to 91%. Cuttings planted in pellets soaked in 100, 250, 500, 1000, and 2000 mg L−1 K-IBA or K-NAA produced ≤ 50% rooting, while those quickly dipped in 2000 mg L−1 K-NAA and planted in hormone-free pellets achieved 83% rooting. Peat pots appear to be a potential alternative to plastic pots, and the laborious work of pot filling can be eliminated by planting cuttings in peat pellets after a quick dip in 2000 mg L−1 K-NAA.

    Seven pomegranate cultivars with excellent fruit quality in eastern Afghanistan were used for hardwood cutting propagation. Cuttings wounded on two opposite sides were treated with 0.5 and 2% Indole-3-butyric acid or 1-Naphthalenacetic acid (NAA) mixed with talc. The auxin treatments induced the same rooting percentage as the control, but NAA 2%, which inhibited shoot growth of cuttings in most of the cultivars, significantly reduced the rooting percentage. Nearly all of the rooted cuttings showed basal rooting, while root development above the wounds, acrobasal rooting, was significantly affected by cultivar and auxin treatment. The cultivars with the highest rooting percentage also had the highest acrobasal rooting percentages. However, the cultivar with the lowest rooting percentage was not the cultivar with the lowest acrobasal rooting percentage. The auxin treatments promoted acrobasal rooting. The type and concentration of auxin that significantly increased the number and length of roots varied among the cultivars.

    Recently, an efficient softwood leaf-bud cutting propagation of persimmon was developed, and indole-3-butyric acid (IBA) was used as an auxin for promoting rooting in almost all the persimmon cutting propagation. We examined the possible use of another auxin, 1-naphthaleneacetic acid (NAA), in cutting propagation of two dwarfing rootstocks for persimmon, 'MKR1' and FDR-1. Using quick-dip method, three concentrations of Na-NAA, water-soluble NAA, were tested for comparing the optimal concentrations of IBA, which were determined in the previous studies. Although the highest concentration treatment damaged some of the cuttings, the treatments to 'MKR1' cuttings in all Na-NAA concentrations tested induced rooting as effectively as the IBA 2000 mg L-1 treatment. The optimal concentration of Na-NAA seemed to be 5500 mg L-1, although there were no significant differences for rooting percentages between the auxin treatments. The rooting percentage of FDR-1 cuttings treated with Na-NAA 11000 mg L-1 as well as with IBA 6000 mg L-1 seemed to be the highest, although there were no significant differences in the auxin-treated cuttings. Between the auxin-treated cuttings, there were no significant differences in the percentages of finished nursery stock, i.e. in the number of overwintering survival of rooted cuttings per the number of original cuttings. Therefore, our study indicated that NAA effectively induced rooting leaf-bud cuttings of dwarfing rootstocks for persimmon as well as IBA did.

    Grafting is the most widely used method for the persimmon clonal propagation, but due to variable agricultural conditions, different skills are adopted by the nurseries across the world. The afghan nursery growers mainly used Diospyros lotus as rootstock for the Diospyros kaki production. A traditional practice is that they cover union of the grafted plant with the mud having a dress of the polyethylene film on the exterior part. Based on the grafters' speculation, mud keeps moisture around the union and through enhances rate of the grafting success. Although no research has been conducted to prove these benefits, this practice is looking laborious, infectious, and time-consuming. The present study was designed to elucidate advantages of the traditional method and assess other useful alternatives for the persimmon grafting in Afghanistan condition. The two-year-old seedlings of D. lotus grown in the field and transplanted to perforated black polyethylene bags at the beginning of their second year were grafted to 'Rojo Brillante' and 'Samarkheli' persimmon cultivars using the cleft method and then subjected to five treatments: covering scion and union with a waterproof paper bag, covering scion and union with a transparent polyethylene bag, sealing scion and union with a layer of buddy tape, sealing graft union with mud, and scion without wrapping or bared. In the second year, the experiment repeated with the same manner, except the ambient (open field and closed polytunnel) was added as a new factor and two previous wrapping treatments (waterproof paper and polyethylene bag) were changed to a food wrapping polyethylene film. After 65 days, percentage of graft success, shoot number, shoot length, and shoot thickness were affected by the treatments. In both years, high graft success (≥92%) achieved with buddy tape while the rest including mud treatment resulted no more than ten percent success. Neither the cultivar nor the ambient had a significant effect over the success of grafting. The 'Rojo Brillante' produced significantly more shoot number, shoot length, and shoot thickness per plant than 'Samarkheli' at both years. Thus, instead of mud, whole scion and union of the persimmons should be sealed with a layer of buddy tape in order to obtain optimum grafting success.

    This study was conducted to obtain insights into unreduced gamete formation in <i>Citrus tamurana</i> hort. ex Tanaka ‘Nishiuchi Konatsu’ and ‘Hyuganatsu’, inferred from unreduced pollen and egg cell genotype information. To establish a method for fine genotyping of tissues with higher ploidy levels (up to hexaploidy), we first identified heterozygous single-nucleotide polymorphism (SNP) markers (CiC4240-04 and CiC5327-03) with close genetic distance to centromeres of ‘Nishiuchi Konatsu’ and ‘Hyuganatsu’. Fluorescence intensity-based standard plots were generated using real-time qPCR to accurately genotype these SNP markers on a two-dimensional panel. Ten normal seeds were obtained from fertilization of unreduced gametes by self-pollination of ‘Nishiuchi Konatsu’ or ‘Hyuganatsu’ × ‘Nishiuchi Konatsu’ crosses. Inner seed coats with hexaploid endosperm tissues and tetraploid embryos were obtained from normal seeds, and their CiC4240-04 and CiC5327-03 genotypes were determined with standard plots using real-time qPCR. Genotypes of unreduced pollen and egg cells were uniquely determined from the genotype information of 6x endosperms and 4x embryos, and the unreduced gamete formation process was inferred. The results showed that unreduced pollen formation was almost entirely due to first division restitution (FDR); therefore, it was concluded that FDR was the dominant formation process for unreduced pollen in ‘Nishiuchi Konatsu’. In contrast, there was no dominant process for the formation of unreduced egg cells because both FDR and second division restitution occurred in almost the same number of individuals with unreduced egg cells.

    研究代表者らが開発したわい性台木MKR1は樹をわい化させるだけでなく、早期着花性、果実高生産性、生理的落果防止など実際栽培に有益な形質を穂木品種に与えることがわかり、実用化へ向けた全国規模の連絡栽培試験が進められている。MKR1台木によるわい化の仕組みは、リンゴ等のわい性台木とは異なることを明らかにしたが、主原因は全く不明であり、今後、栽培指針を示し、新規わい性台木を効率的に育種するための指標や分子マーカー等を開発するためには、その主原因を明らかにしなければならない。最近、サイトカイニン含量を高めた形質転換果樹がわい化成長することが示されたが、MKR1台木樹はその形質転換体と非常に似た成長を示している。そこで本研究ではMKR1台木の成長制御機構を、サイトカイニンの分析や組換え体の作出、遺伝子発現等を通じて明らかにする。

    植物ペプチドホルモンに関する基礎的研究は、近年めざましい発展を遂げているものの、実用化を目指した本格的な実験はほとんど行われておらず、本研究はその実用化を最終目的としているものである。現在、確認されている15 以上の植物ペプチドホルモンの中から、我々がブドウ等の果樹において関連遺伝子の存在と試験管内処理効果を見いだしたCLE ペプチドを用い、園芸作物への分裂組織(メリステム)の形成制御機構を明らかにする。また、ペプチドホルモン研究の第一人者より最新情報を得ながら、気孔数を調節するストマジェンなどの新規ペプチドホルモンの外生処理や遺伝子の発現等の調査も進める。これらの実験の結果、実用化に結びつくようなデータが得られれば、実際に圃場における処理を試み、実用化に向けた展開を図る。
