油糧生産微生物ラビリンチュラの培養細胞からオイル抽出した残渣をタンパク源として、養魚用の配合飼料を作成した。配合飼料で羽生されている魚粉を用いた試料も同様に作成し、コイに対する飼育試験を実施した。その結果、ラビリンチュラ抽出残渣は魚粉で作成した飼料と同等の成長を示した。

Recently, Dr. Katsuhisa Uchida who belong to University of Miyazaki, Japan, succeeded in establishing a production technique for giant masu salmon by transferring landlocked yamame to the sea for rearing during the winter season. The production rate and yield of the fish-mass significantly increased by culturing in seawater. The purpose of this study was to produce a fish sauce from newly developed fishery source, giant mass salmon by rapid fermentation under no-salt conditions. The whole body of giant masu salmon except for eggs was minced well, and the activity of protease contained in the mixtures was analyzed at 55oC and 60oC according to the adequate temperature indicated by our previous study. The protease activity was comparable to our previous study, and the activity at 55 and 60oC was 33 and 49 Unit/ g-protein, respectively. Giant masu salmon mixtures were incubated to induce autolysis at 55 or 60oC for 72 hrs. During incubation, to evaluate the degree of autolysis, the amino acid composition was analyzed with liquid chromatograph-mass spectrometry (LC-MS). The distribution of protein molecular size was analyzed by SDS-PAGE. The content of total amino acids was increased as time passed, and the low molecular weight bands appeared by incubation. Giant masu salmon mixtures were incubated at 60oC for 24 h, and the centrifugal supernatant was heated in boiling water for 20 min. The supernatant of the heated solution was used as giant masu salmon fish sauce (GMSFS). The ratio of anserine which well known as a functional amino acid, in GMSFS was significantly high (≒48% to total amino acid). Thus, the anserine-rich fish sauce could be produced quickly from giant masu salmon. At present, to add more functionality to GMSFA and modify the taste, we are carrying out the fermentation trial of GMSFS with lactic acid bacteria after autolysis.

By previous our study, Lactococcus lactis strain K-C2 showed antagonistic activities against various pathogens in aquaculture such as Edwardsiella tarda, Lactococcus garviae and Streptococcus iniae. Therefore, this strain is considered as novel probiotics to prevent disease in aquaculture. In this study, we tried to enrich L. lactis strain K-C2 in the feed organisms, Artemia sp. Commercial eggs of Artemia (Pacific Trading Co. Ltd., Fukuoka, Japan) were hatched in an artificial seawater under the vigorous aeration and light irradiation. The Artemia after 4 days of hatching was introduced to 80 ml of artificial seawater in a flask (26 individuals/ml). Strain K-C2 was added to the seawater containing Artemia at 3.2×108 cfu/ml. As a control group, Artemia was cultured without addition of strain K-C2. After exposure to the cell suspensions of strain K-C2, Artemia was harvested and washed by sterile artificial seawater and homogenized. The homogenates was serially diluted, and each dilutions were inoculated on a GYP agar plate medium containing calcium carbonate. The plates were incubated at 28oC for 4 days and viable colonies were counted to calculate colony forming unit (CFU). The colonies which degraded the calcium carbonate were randomly selected and colony PCR was carried out using Lactococcus specific primer set (Forward primer, LacF 5’ GTACTTGTACCGACTGGAT 3’ and reverse primer, LacreR 5’ GGGATCATCTTTGAGTGAT 3’) to detect the strain K-C2. The selected colonies were estimated as L. lactis by PCR with specific primers. The number of L. lactis in Artemia was calculated to 4.2×103 CFU/ 100 individuals. This study indicated that probiotic L. lactis strain K-C2 could be enriched in Artemia by exposure in seawater.
