Widely distributed from East Asia to Australia, Rubus parvifolius is the only known wild Rubus species that naturally grows throughout the Japanese Archipelago, including small southern islands. R. parvifolius has been attracting attention as a genetic resource in Asia for breeding raspberries with enhanced tolerance to environmental stresses. Amami Oshima and Kakeroma, two of Japan’s Amami Islands, exhibit high plant diversity due to significant species endemism, reflecting the islands' isolated biogeographical history. This provides a rationale for focusing on Amami Oshima and Kakeroma Islands as a suitable system for studying wild Rubus species. In this study, we conducted ploidy assessments and analyzed Simple Sequence Repeat (SSR) markers in 24 accessions of R. parvifolius and other wild Rubus species (R. grayanus and R. croceacanthus) from those islands. Ploidy evaluation using flow cytometry and chromosome observation revealed that 11 of the 24 accessions (45.8%) were triploid. In addition, 52 SSR markers were employed, of which 20 were selected for analysis. Principal coordinate analysis (PCoA) using these SSR markers showed that diploid and triploid individuals did not form distinct clusters. Although this study was preliminary and based on a limited sample size, a relatively frequent occurrence of polyploid individuals was observed among the R. parvifolius accessions examined from Amami Oshima and Kakeroma. Our results provide baseline information on the occurrence of polyploid individuals in wild species in island systems.

Foot rot, a disease caused by the fungal plant pathogen Diaporthe destruens, has been a major problem throughout East Asia. In major sweet-potato-producing regions, developing sweet potato cultivars that are resistant to foot rot has become an urgent priority. The possibility of selecting resistant cultivars by using polyphenols in the stems as markers was recently suggested, but this selection method has not been tested in the crossbreeding of sweet potato cultivars. In this study, we crossed the sweet potato cultivars ‘Konaishin’ and ‘Tamaakane’ (each of which is resistant to foot rot), analyzed the polyphenols in the stems of the resulting hybrid lines, and evaluated the possibility of selecting resistant lines. As a result, KT No. 7 and KT No. 8 showed similar or lower total polyphenol contents (145.9 and 112.9 mg GAE 100 g−1 FW, respectively) compared to ‘Tamaakane’ (142.2 mg GAE 100 g−1 FW). The selected line, KT No. 7, exhibited the highest resistance among the hybrids when it was directly inoculated with the foot rot pathogen using stems as test material, showing a disease severity value of 1.8, which was substantially lower than that of ‘Tamaakane’ (50.0). These results suggest that stem polyphenol content has potential as a marker for identifying promising candidates with foot rot resistance, although its predictive value may vary depending on genetic and environmental factors. This approach may help improve the efficiency of foot rot resistance screening in sweet potato breeding programs.ms.

Foot rot (Diaporthe destruens) of sweetpotato generally infects the stem of the plant and then spreads throughout the plant. In Japan, foot rot is prevalent in the main area of sweetpotato production, and there is a pressing need for the development of resistant cultivars. It is generally believed that polyphenols are involved in defense against stress. However, little is known about the relationship between polyphenols and the susceptibility to foot rot in sweetpotato. We thus analyzed the polyphenol of the disease-free healthy stems and investigated their relationship between susceptibility to the disease to establish basic techniques for selecting cultivars resistant to foot rot. Polyphenol was analyzed by the Folin–Ciocalteu method and high-performance liquid chromatography. Resistance tests were conducted using the direct inoculation method. We observed a correlation between the total polyphenol content and the degree of foot rot disease severity. Surprisingly, cultivars with a low stem polyphenol content (‘Tamaakane’) had lower susceptibility to foot rot, whereas the cultivars with high stem polyphenol content (‘Kokei No. 14’) had higher susceptibility to foot rot. In particular, high positive correlations were shown with several polyphenols, including 3,4-dicaffeoylquinic acid and chlorogenic acid. Furthermore, the results of in vitro experiments also suggested that foot rot may metabolize trace amounts of chlorogenic acid. This discovery has potential applications in the selection of resistant cultivars, contributing to the streamlining of disease resistance tests in breeding programs. It could ultimately shorten the breeding period and help overcome foot rot disease in sweetpotato.

Key message: In Cyrtanthus mackenii, development of embryo and endosperm were differentially affected by fertilization of male gametes with DNA damage and mutations. Abstract: Pollen irradiation with ionizing radiations has been applied in plant breeding and genetic research, and haploid plant induction has mainly been performed by male inactivation with high-dose irradiation. However, the fertilization process of irradiated male gametes and the early development of embryo and endosperm have not received much attention. Heavy-ion beams, a type of radiation, have been widely applied as effective mutagens for plants and show a high mutation rate even at low-dose irradiation. In this study, we analyzed the effects of male gametes of Cyrtanthus mackenii irradiated with a carbon-ion beam at low doses on fertilization. In immature seeds derived from the pollination of irradiated pollen grains, two types of embryo sacs were observed: embryo sac with a normally developed embryo and endosperm and embryo sac with an egg cell or an undivided zygote and an endosperm. Abnormalities in chromosome segregation, such as chromosomal bridges, were observed only in the endosperm nuclei, irrespective of the presence or absence of embryogenesis. Therefore, in Cyrtanthus, embryogenesis is strongly affected by DNA damage or mutations in male gametes. Moreover, various DNA contents were detected in the embryo and endosperm nuclei, and endoreduplication may have occurred in the endosperm nuclei. As carbon-ion irradiation causes chromosomal rearrangements even at low doses, pollen irradiation can be an interesting tool for studying double fertilization and mutation heritability.

The leaves and branches of rabbiteye blueberry are rich in proanthocyanidins, which are thought to have different physiological activities depending on their structure and degree of polymerization. In this study, we analyzed the constituents of the leaves and branches of rabbiteye blueberry to determine the seasonal variations in polyphenol and proanthocyanidin (PAC) contents as well as their mean degrees of polymerization (mDP). Total PAC content was determined using two methods: The p-dimethylaminocinnamaldehyde (DMACA) method, which measures monomeric PAC, showed an increase from spring to summer in both leaves and branches. On the other hand, using the butanol/HCl method, which measures only polymerized PAC, the PAC content of leaves increased from spring to summer but those of branches remained low throughout the year, showing no significant increase or decrease. Furthermore, analysis of the mDP of PAC showed increases from spring to summer in the leaves of ‘Kunisato 35 gou’. Although the highest value (8.0) was observed in October, values around 4 remained throughout the year in the branches. Since differences in polymerization degree affect absorption in the body and physiological properties such as antioxidant capacity, selecting the appropriate harvest time and plant organs for each purpose is expected to ensure the quality of processed blueberry foods.

Heavy-ion beam, a type of ionizing radiation, has been applied to plant breeding as a powerful mutagen and is a promising tool to induce large deletions and chromosomal rearrangements. The effectiveness of heavy-ion irradiation can be explained by linear energy transfer (LET; keV µm-1). Heavy-ion beams with different LET values induce different types and sizes of mutations. It has been suggested that deletion size increases with increasing LET value, and complex chromosomal rearrangements are induced in higher LET radiations. In this study, we mapped heavy-ion beam-induced deletions detected in Arabidopsis mutants to its genome. We revealed that deletion sizes were similar between different LETs (100 to 290 keV μm-1), that their upper limit was affected by the distribution of essential genes, and that the detected chromosomal rearrangements avoid disrupting the essential genes. We also focused on tandemly arrayed genes (TAGs), where two or more homologous genes are adjacent to one another in the genome. Our results suggested that 100 keV µm-1 of LET is enough to disrupt TAGs and that the distribution of essential genes strongly affects the heritability of mutations overlapping them. Our results provide a genomic view of large deletion inductions in the Arabidopsis genome.

Sweet potato is a widely cultivated crop with storage roots. Although many studies have been conducted on the mechanism of its storage root formation, the details have not been fully elucidated. We screened mutant lines with inhibition of storage root formation to clarify parts of the mechanism. In this study, the process of storage root formation in one of the mutant lines, C20-8-1, was investigated. The inhibition of storage root formation was observed during the early stages of growth. The roots in C20-8-1 did not show histological differences compared to those in wild type. The transition from fibrous roots to pencil roots, which are the developmental stages prior to mature storage root formation, was delayed or inhibited in C20-8-1. The upregulation of starch biosynthesis-related genes and downregulation of lignin biosynthesis genes with storage root swelling were not confirmed in the root of C20-8-1 during the developmental transition stage, suggesting that most of the roots in C20-8-1 are in the pre-transition state toward the storage root swelling. C20-8-1 showed a mutant phenotype during the critical period of storage root swelling initiation, and further clarification of this mutation is expected to provide new insights into storage root formation.

Sweet potato is a major root crop with nutritious tuberous roots. The mechanism of tuberous root development has not yet been adequately elucidated. Genetic resources are required to develop the molecular understanding of sweet potato. Heavy-ion beams were applied to hexaploid sweet potato for an increase in genetic variation, after which the comprehensive effects of heavy-ion beam irradiation were investigated. In vitro cultured shoots with an axillary bud of ‘Beniharuka’ were irradiated with Ar-ions at a dose of 1–5 Gy and C-ions at a dose of 5–20 Gy, and three irradiated lines were separated from each irradiated shoot. The shoot regeneration was inhibited at high doses of each ion irradiation. Ar-ion irradiation had an especially high biological effect on shoot regeneration. A total of 335 lines were obtained, consisting of 104 and 231 lines derived from Ar- and C-ion irradiation, respectively. The change in the DNA content of the lines was analyzed by flow cytometry to evaluate the irradiation-induced damage to the DNA. The two lines demonstrated significant differences in the DNA content and changes at the chromosome level. The screening for the morphological mutants was conducted in the field. Some irradiated lines showed inhibited or no tuberous root phenotype as mutant candidates. Additionally, the high-yield mutant candidates were dominated by Ar-ion irradiation. It was indicated that heavy-ion beam mutagenesis is effective in broadening the range of the phenotypes corresponding to tuberous root formation in hexaploid sweet potato.

Anthocyanins in sweetpotato (Ipomoea batatas L.) have attracted much attention in light of their multiple health benefits and excellent properties as processed foods. Although crossbreeding is an effective way to breed new cultivars of sweetpotato, there are few reports on how anthocyanin content and composition are passed on to F1 hybrid generations by crossbreeding because of a complex genome and self-incompatibility reactions. In this study, three purple-fleshed sweetpotao cultivars, ‘Akemurasaki’, ‘Kyushu No.137’, ‘Purple Sweet Lord’, and one yellow-fleshed cultivars, ‘HE306’ (Silk Sweet®) selected for their anthocyanin contents and compositions were crossed in six combinations. Flowering was induced by using a sweetpotato grafting technique to obtain a total of 143 F1 hybrid lines. Anthocyanin content, the percentage of acylated anthocyanins, and the peonidin/cyanidin ratio in each of these F1 hybrid lines were investigated. In the F1 hybrid generations derived from these six cross combinations, many lines tended to have anthocyanin contents similar to those of the parent cultivars. In particular, among the 36 hybrid lines derived from crosses between ‘Akemurasaki’ and ‘HE306’ (Silk Sweet®), 13 lines with high anthocyanin contents (≧200 mg·100 g FW−1) and 20 lines without anthocyanins were identified. The F1 hybrid lines of this combination had anthocyanin contents similar to those of the parent cultivars, with anthocyanin-free lines appearing more frequently than anthocyanin-containing lines. Interestingly, we also identified a line with an extremely low percentage of acylated anthocyanins (49.7%) from the F1 hybrid lines derived from a cross between ‘Purple Sweet Lord’ (seed parent) and ‘Kyushu No. 137’ (pollen parent). Moreover, the F1 hybrid lines derived from crosses between ‘Purple Sweet Lord’ and ‘Kyushu No.137’ tended to have high peonidin/cyanidin ratios. This study provides an example of the inheritance of anthocyanin content and its composition in sweetpotato, which supplies valuable information for selection of parents to breed purple-fleshed cultivars.

DNA Damage Response of Cyrtanthus mackenii Male Gametes Following Argon Ion Beam Irradiation 査読あり

Heavy-ion beams have been applied as effective mutagens to various plant materials. Pollen has been used as material for mutant induction and genetic analysis. However, our knowledge of the DNA damage response of plant male gametes remains limited. In the present study, we irradiated <i>Cyrtanthus mackenii</i> pollen with an argon ion beam, which induced complex DNA damage, and investigated the DNA damage response of male gametes during pollen tube growth. Male gametes derived from the irradiated pollen grains were isolated from pollen tubes after 12 and 24 h of culture and subjected to cell cycle analysis. After 12 h of culture, the irradiated generative cells were frequently arrested at metaphase during pollen mitosis II (PMII), and the proportion of metaphase cells increased with increasing absorbed dose. These results suggest that the genomic lesions induced by the argon ion beam caused spindle assembly checkpoint (SAC)-dependent arrest. After 24 h of culture, the irradiated male gametes completed PMII, albeit forming sperm cells with abnormalities in chromosome separation, and chromosomal bridges were often formed between these cells. Moreover, phosphorylated H2AX foci, an indicator of DNA double-strand breaks, were detected in the irradiated male gametes after 24 h of culture, regardless of passing through the SAC. Taken together, these results indicate that male gametes activate functions to cope with radiation-induced complex DNA damage during pollen tube growth.

In <i>Undaria pinnatifida</i>, an effective method for mutant screening in sporophytes has not been established. The present study developed a novel mutant screening method for <i>Undaria</i> sporophyte by combining gametophyte mutagenesis with heavy-ion beam and land-based tank culture system. When we irradiated gametophytes and sporophytes with carbon- and argon-ion beams, survival rates of the female gametophytes and the sporophytes decreased with increasing dose. However, those of the male gametophyte did not decrease after both of the irradiations. Mutant screening during the sporophyte development was performed by using a land-based tank culture system. High-growth plants were selected in the first mutant (M<sub>1</sub>) population derived from the irradiated materials. We successfully obtained mutant candidates with higher growth than the wild type in the M<sub>2</sub> generation obtained from brother-sister inbreeding of selected M<sub>1</sub> plants. Four high-growth mutant candidate lines were selected from M<sub>2</sub> populations of 48 lines. The mutant candidates were derived from 3 lines of the gametophyte irradiation and 1 line of the sporophyte irradiation, suggesting that the materials for the irradiation are applicable for mutant induction. The mutant screening method and the selected mutant candidates would advance the breeding and molecular biology in <i>U. pinnatifida</i>.

BACKGROUND: In blueberries, the total polyphenol contents, total anthocyanin contents, and antioxidant activities of fruits in blueberries and their relatives are becoming important targets for breeders. Recently, intersectional hybrids between Japanese wild species Shashanbo and highbush blueberry were produced using polyploid breeding for the first time. However, the polyphenol contents, antioxidant activities, and anti-cancer cell proliferation properties of the fruits of these intersectional hybrids have not been studied sufficiently. OBJECTIVE: The objectives of this study were to determine the anthocyanin contents, polyphenol contents, and antioxidant activities at each stage of fruit development in intersectional hybrids, and to examine the effects of the fruit extracts on anti-cancer cell proliferation properties using human promyelocytic leukemia (HL-60) cells. METHODS: Freeze-dried fruit samples of three intersectional hybrids and their parents at five developmental stages were used to evaluate their polyphenol contents, anthocyanin contents, and levels of two types of antioxidant activities (DPPH and ORAC). The anti-cancer cell proliferation properties of each extract were also evaluated according to the survival rate of HL-60 cells. RESULTS: The total polyphenol contents in fruits of the intersectional hybrids gradually decreased as the fruits grew, but the contents increased slightly at the mature stage. Meanwhile, the total anthocyanin contents increased strongly to the mature stage. In addition, the fruit extract of intersectional hybrid JM1 was found to be more effective for inhibiting the growth of HL-60 human leukemia cells in vitro. Total polyphenol content correlated highly with antioxidant activities and anti-cancer cell proliferation properties. CONCLUSIONS: We clarified that the intersectional hybrids had produced have abundant bioactive compounds, high levels of antioxidant activities, and anti-cancer cell proliferation properties. The results proved the effectiveness of the use of Japanese wild Vaccinium species for the strategic breeding of blueberries with high levels of bioactive compounds.

Two growth processes, cell proliferation and expansion, determine plant species-specific organ sizes. A large flower mutant in Arabidopsis thaliana, ohbana1 (ohb1), was isolated from a mutant library. In the ohb1 flowers, post-mitotic cell expansion and endoreduplication of nuclear DNA were promoted. The whole-genome resequencing and genetic analysis results showed that the loss of function in MEDIATOR16 (MED16), a mediator complex subunit, was responsible for the large flower phenotypes exhibited by ohb1. A phenotypic analysis of the mutant alleles in MED16 and the double mutants created by crossing ohb1 with representative large flower mutants revealed that MED16 and MED25 share part of the negative petal size regulatory pathways. Furthermore, the double mutant analyses suggested that there were genetically independent pathways leading to cell size restrictions in the floral organs which were not related to the MED complex. Several double mutants also formed larger and heavier seeds than the wild type and single mutant plants, which indicated that MED16 was involved in seed size regulation. This study has revealed part of the size-regulatory network in flowers and seeds through analysis of the ohb1 mutant, and that the size-regulation pathways are partially different between floral organs and seeds.

The breeding of raspberries (Rubus idaeus L.) and blackberries (R. fruticosus Agg.) has shifted in focus toward traits associated with the sensory qualities and potential health benefits of the fruit, and Rubus wild species have attracted a great deal of attention as new sources of genetic diversity. In this study, the anthocyanin and polyphenol contents and compositions, and three types of antioxidant capacities were compared in mature fruits including raspberries, blackberries, and Japanese wild Rubus species. ‘Blackcap’ black raspberry showed the significantly highest values of total anthocyanin content and total polyphenol content among 12 Rubus species or cultivars. Cyanidin-3-xylosyl-rutinoside was contained only in black raspberries, and was characteristic anthocyanin in Rubus species. R. croceacanthus H. Lév., a Japanese wild species that is adapted to warm regions, had a deep red fruit color, and had the highest total anthocyanin and total polyphenol contents among the four Japanese wild species examined. As for the antioxidant capacities, the highest values were observed in ‘Blackcap’ among 12 Rubus species. Furthermore, a high positive correlation was shown between total anthocyanin or total polyphenol content and antioxidant capacities. In particular, black raspberries and blackberries contained high concentrations of rutin. Thus, rutin might contribute to the high antioxidant capacities in these cultivars.

The genetic analysis of five haploid plants (BX1-BX5) of ‘Banpeiyu’ pummelo (BP), [Citrus maxima (Burm.) Merr.] using 32 simple sequence repeat (SSR) markers carried out for the purpose of confirming their gynogenetic origin and genetic differences. As the results, all the haploid plants possessed only either one of the BP alleles, confirming that they are gynogenetic origin. We also confirmed that BX1 and its doubled haploid (BX1-DH) have exactly the same SSR alleles. Interestingly, all the five haploids had common alleles on the linkage group (LG) 6 in the case of three markers. Then, we performed self-pollination of DH plants and reciprocal crosses between DH and BP, which has strong self-incompatibility. The seeds obtained by these crosses germinated in vitro or ex vitro normally and developed into diploid seedlings, except for one tetraploid seedling. Genetic analysis of these seedlings with 32 SSR markers revealed that segregation ratios of the 32 loci in the 30 BP inbred, 30 BP x DH and 30 DH x BP progenies mostly fitted to the expected ratios. However, significantly distorted segregation ratios were found in one, five and three SSR markers for BP inbred, BP x DH and DH x BP progenies, respectively. All of the 30 DH inbred progenies showed exactly the same SSR alleles as the original DH. This is the first example in which self and back cross progenies of DH in fruit crops were genetically analyzed using SSR markers.

© 2019 Japanese Society of Phycology Undaria pinnatifida, the brown macroalga, is a major commercial edible seaweed, and there is increasing interest in breeding new, improved cultivars for market expansion. In this study, we attempted to establish mutagenesis in U. pinnatifida using Ar and C ion beams as mutagens to meet future demands. To screen irradiated generations for mutants, U. pinnatifida zoospores irradiated with Ar and C ion beams were cultivated in plastic Petri dishes. Some gametophytes derived from the irradiated zoospores showed growth arrest or cell death at the initial developmental stage. Although the growth inhibition and lethal effects were observed at high doses of each ion irradiation, the Ar ion irradiation had high biological effects on cell division and growth. The gametophytes that showed a reduction in cell elongation were designated as an inhibited cell elongation mutant. A comparison of the mutant induction frequencies revealed that the C ion beam showed a higher frequency than the Ar ion beam. The highest frequency was 0.83% at 12.5 Gy of the C ion beam. We determined the total number of sporophytes and embryos per female gametophyte after sporophyte induction. High-dose irradiation with the Ar ion beam decreased the embryo and sporophyte formation, suggesting that the Ar ion beam also has exhibited high biological effects on the fertilization or embryogenesis processes or both. The developed heavy ion mutagenesis and mutant screening methods would be useful for mutation breeding and constructing specific mutant libraries in brown algae, and not only in U. pinnatifida.

The irradiation treatment is quick, lasting between a few seconds and a few minutes, and is sufficient to induce mutation. Some new cherry blossom cultivars have already been introduced to the market. Pale-yellow cherry blossom 'Nishina Zao' was created by irradiating greenish 'Gyoiko' (Prunus lannesiana) scions with carbon ions. Unlike normal cherry blossom trees (P. pauciflora 'Keio-zakura Yamagata No.13'), 'Nishina Otome' does not require a certain period under cold winter weather to induce flowering in the spring. Both cultivars are propagated by cloning branches of mutant plants. We have also succeeded in creating new cultivars using progeny of irradiated plants that are not changed with respect to either flower color or shape. Although the original cultivar, P lannesiana 'Beni-yutaka', has a double flower with 15-20 petals, the new cultivar 'Nishina Tomoka' has a large, single flower with 5 petals. Heavy-ion beams are thus effective tools for creating new variations of Prunus trees through mutation breeding.

Chromosomal rearrangements are thought to play a role in speciation, adaptation and evolution. The effects of chromosomal rearrangements on gene expression have been reported in many species and their close relatives, and may illustrate the consequences of adaptation. To investigate the effects of chromosomal rearrangements on phenotypic changes directly, studies of experimentally induced rearrangements are required; they allow gene expression and epigenetic states to be compared between mutants and wild-types. We have developed an effective method for inducing chromosomal rearrangements with heavy-ion beam irradiation, which is a sort of ionizing radiation. Reconstruction of such rearrangements by genome editing will enable reproducible studies on the effects of chromosomal rearrangements. In the future, therefore, these techniques will usher in a new era in which researchers can create novel species with designer chromosomes.

Several studies have indicated that the value of LET affects DNA lesion formation in several ways, including the efficiency and the density of double-stranded break induction along the particle path. We assumed that the mutation type can be altered by selecting an appropriate LET value. Here, we quantitatively demonstrate differences in the mutation type induced by irradiation with two representative ions, Ar ions (LET: 290 keV μm −1 ) and C ions (LET: 30.0 keV μm −1 ), by whole-genome resequencing of the Arabidopsis mutants produced by these irradiations. Ar ions caused chromosomal rearrangements or large deletions (≥100 bp) more frequently than C ions, with 10.2 and 2.3 per mutant genome under Ar- and C-ion irradiation, respectively. Conversely, C ions induced more single-base substitutions and small indels ( < 100 bp) than Ar ions, with 28.1 and 56.9 per mutant genome under Ar- and C-ion irradiation, respectively. Moreover, the rearrangements induced by Ar-ion irradiation were more complex than those induced by C-ion irradiation, and tended to accompany single base substitutions or small indels located close by. In conjunction with the detection of causative genes through high-throughput sequencing, selective irradiation by beams with different effects will be a powerful tool for forward genetics as well as studies on chromosomal rearrangements.

Undaria pinnatifida is grown for food and industrial materials worldwide; therefore, advanced breeding is needed to meet quality and productivity requirements. In this study, we examined regional lines of U. pinnatifida from five cultivation sites in Japan with different environmental conditions: Oga (OGA, the northern Sea of Japan coast), Hirota Bay (HRT, the northeastern Pacific coast), Matsushima Bay (MAT, the northeastern Pacific coast), Naruto (the Seto Inland Sea coast) and Shimonoseki (SIM, the southern Sea of Japan coast). The sporophytes of these lines were cultured in a tank culture system under controlled environmental conditions, and their morphological characteristics, nutrient uptake kinetics (V max , K s and V max /K s ), and carbon, nitrogen and phosphorus contents were determined. Sporophytes from MAT grew faster, whereas those from SIM were smaller than those from the other sites. Although the blade thickness of sporophytes cultivated in the sea significantly differs among cultivation sites in the previous study, there was no significant difference in blade thickness among the regional lines cultivated in the tank. Sporophytes from OGA had the greatest V max /K s values and significantly greater nitrogen contents than the other lines. Therefore, the morphological characteristics of MAT and SIM sporophytes, and the nutrient uptake kinetics of OGA sporophytes may have a genetic origin. This indicates that these lines may represent useful resources for selective breeding, with MAT sporophytes providing faster growth and OGA sporophytes being well-adapted to low-nutrient conditions.

© 2016, Springer Science+Business Media Dordrecht. Kelps are economically valuable primary producers; therefore, many studies on breeding have attempted to increase kelp productivity and quality. However, most cultivation tests have been performed in the ocean, thereby limiting the development of new cultivars. To reduce the breeding duration period and confirm cultivar phenotypes, we developed a novel tank culture system, referred to as a circulation and floating culture system (CFCS), for cultivating macroalgae. In the CFCS, kelp can be cultivated under controlled environmental conditions. Water velocity in the CFCS can be regulated by changing the angle of a seawater inlet spout without changing the volume of seawater in the tank. Undaria pinnatifida and Saccharina japonica cultivated in the CFCS exhibited morphological features very similar to those of plants grown naturally in the ocean. The result suggests that the facility is useful for identifying water motion conditions suitable for increasing the production of any macroalgae species. Using this facility, both species were grown from juvenile sporophytes (20 mm) to maturity; for U. pinnatifida, the subsequent g eneration was successfully cultivated. Improved growth of U. pinnatifida was achieved in fast flows compared with slow flows, whereas S. japonica developed a wider shape and heavier biomass in slow flows compared with fast flows. We discuss the application and implication of the CFCS for breeding research and the physiological ecology of macroalgae.

Understanding the nutrient uptake kinetics of kelp populations will contribute to an improved understanding of environmental adaptation and the breeding of new cultivars. In this study, we examined the morphological characteristics, carbon (C) and nitrogen (N) contents, and NO<inf>3</inf> <sup>−</sup>–N and NH<inf>4</inf> <sup>+</sup>–N uptake kinetics of Undaria pinnatifida sporophytes cultivated at six industrial farms throughout Japan. We detected significant differences in morphology among sites. At Matsushima Bay (northern Pacific coast of Japan), where autumnal seawater temperatures fall in concert with increasing nutrient concentrations, sporophytes were significantly larger than at other sites from December to February. The C content of the sporophytes was seasonally stable at all of the locations, but the N content of sporophytes declined after February due to a decrease in seawater nutrients. We compared the uptake kinetics of NO<inf>3</inf> <sup>−</sup>–N and NH<inf>4</inf> <sup>+</sup>–N among cultivation sites. V<inf>max</inf> and K<inf>s</inf>, which are Michaelis–Menten parameters that measure adaptation to nutrient concentrations, were highest in the Seto Inland Sea and lowest in the northern sector of the Sea of Japan. The V<inf>max</inf>/K<inf>s</inf> ratio is a measure of adaptation to low nutrient concentrations; the highest values were measured in the northern sector of the Sea of Japan. The parameter ranges were broader than those previously reported for invasive populations of U. pinnatifida in other parts of the world. Thus, we detected population-level adaptations to the various nutrient conditions in Japanese waters, and these results suggest the existence of ecotypes according to nutrient uptake kinetics. The different populations may be used to provide sources of genetic material that could be of value in breeding programmes by improving productivity and quality.

The RIKEN RI Beam Factory (RIBF) is the one of the biggest facilities to accelerate heavy ions in the world. Nuclear physics is the primary subject of the RIBF. You may know RIKEN as nuclear physics laboratories. Its activities cover every scientific and technological fields, even in agriculture. We have developed a unique technology for mutation induction by using heavy-ion beams from particle accelerators at the RIBE, through an efficient synergistic link between agricultural science and accelerator physics. At relatively low doses, ion beams induce mutations at a high rate without severely inhibiting growth. The irradiation treatment given to the various plant materials is quick, lasting between a few seconds and a few minutes, but is sufficient to induce mutation. By using this method, we already put 26 new cultivars on the market in Japan, the United States, Canada, and the European Union since 2001. For example, rose induced mutations are utilized in vegetative propagated crops to shorten the time necessary to breed for desirable characters (Figure 1). In the present article, we introduce the development and practical applications of ion-beam breeding, effects of Linear Energy Transfer (LET) on lethality and mutagenesis, and future perspectives on biological applications of heavy-ion beams.

Self-incompatibility (SI) is an important mechanism in higher plants that promotes outcrossing and prevents self-fertilization. ‘Banpeiyu’ (Citrus maxima) and ‘Hyuganatsu’ (Citrus tamurana), two of the Citrus cultivars distributed in Kyusyu, Japan, show gametophytic SI. In this study, we used the Citrus mature pollen culture system and stylar crude protein extracts to simulate compatible (C) and SI responses in ‘Banpeiyu’ pollen tubes. We analyzed the protein changes in pollen tubes with the C- and SI-like treatments by nano-liquid chromatography-mass spectrometry (nano-LC-MS); 14 and 27 proteins were identified in C- and SI-like treatments, respectively. We picked up some candidate genes that were particularly prevalent in SI-like treatment and analyzed their expression level changes during C- and SI-like treatments in ‘Banpeiyu’ and ‘Hyuganatsu’ pollen tubes. The expression levels of copper/zinc superoxide dismutase (Cu/Zn SOD), manganese SOD (Mn SOD), catalase (CAT), and cysteine protease (CYP) increased after SI-like treatment. We used a fluorescent probe to visualize reactive oxygen species (ROS) level changes in ‘Banpeiyu’ and ‘Hyuganatsu’ pollen tubes after C- and SI-like treatments and found that 2-hour SI-like treatment induced ROS levels to increase in the pollen tubes of both cultivars. These results suggest that an ROS increase could be one of the key phenomena in the SI response of Citrus and that gene expression changes were responses to ROS generation.

Heavy-ion beams are widely used for mutation breeding and molecular biology. Although the mutagenic effects of heavy-ion beam irradiation have been characterized by sequence analysis of some restricted chromosomal regions or loci, there have been no evaluations at the whole-genome level or of the detailed genomic rearrangements in the mutant genomes. In this study, using array comparative genomic hybridization (array-CGH) and resequencing, we comprehensively characterized the mutations in Arabidopsis thaliana genomes irradiated with Ar or Fe ions. We subsequently used this information to investigate the mutagenic effects of the heavy-ion beams. Array-CGH demonstrated that the average number of deleted areas per genome were 1.9 and 3.7 following Ar-ion and Fe-ion irradiation, respectively, with deletion sizes ranging from 149 to 602 180 bp; 81% of the deletions were accompanied by genomic rearrangements. To provide a further detailed analysis, the genomes of the mutants induced by Ar-ion beam irradiation were resequenced, and total mutations, including base substitutions, duplications, in/dels, inversions, and translocations, were detected using three algorithms. All three resequenced mutants had genomic rearrangements. Of the 22 DNA fragments that contributed to the rearrangements, 19 fragments were responsible for the intrachromosomal rearrangements, and multiple rearrangements were formed in the localized regions of the chromosomes. The interchromosomal rearrangements were detected in the multiply rearranged regions. These results indicate that the heavy-ion beams led to clustered DNA damage in the chromosome, and that they have great potential to induce complicated intrachromosomal rearrangements. Heavy-ion beams will prove useful as unique mutagens for plant breeding and the establishment of mutant lines. Significance Statement Heavy-ion beams, a form of ionizing radiation, efficiently induced both large deletion mutations and genomic rearrangements. The heavy-ion beams led to clustered DNA damage in the chromosome, and that they have a great potential to induce complicated intrachromosomal rearrangements.

Citrus is a genus with gametophytic self-incompatibility, which prevents fertilization by self-pollination. Polyamines (PAs), as a class of active small molecules, widely participate in various cellular activities. To investigate the effects of the changes in PA contents [putrescine, spermidine (SPD), and spermine] on pollen tube growth in self-incompatible (SI) plants, we cultured pollen of Citrus maxima ‘Banpeiyu’ and C. tamurana ‘Hyuganatsu’ in a Citrus mature pollen culture system and detected the PA content changes in pollen by high-performance liquid chromatography (HPLC) analysis during germination and after treatment with crude protein extracts of compatible or incompatible styles. We profiled the PA content changes during the germination course in pollen of ‘Banpeiyu’ and ‘Hyuganatsu’ by HPLC, and identified increases of free and perchloric-acid-insoluble SPD contents in pollen tubes in the incompatible treatment. Exogenous SPD treatment obviously increased the SPD contents in the pollen tube, inhibited pollen tube elongation, and resulted in some abnormal morphological alterations, such as tip inflation, tube content leakage and tip twist, which were observed in both ‘Banpeiyu’ and ‘Hyuganatsu’ pollen tubes exposed to incompatible treatment. This finding implied that SPD might participate in the SI response in Citrus.

Plant shoot organs such as stems, leaves and flowers are derived from specialized groups of stem cells organized at the shoot apical meristem (SAM). Organogenesis involves two major processes, namely cell proliferation and differentiation, whereby the former contributes to increasing the cell number and the latter involves substantial increases in cell volume through cell expansion. Co-ordination between the above processes in time and space is essential for proper organogenesis. To identify regulatory factors involved in proper organogenesis, heavy-ion beam-irradiated de-etiolated (det) 3-1 seeds have been used to identify striking phenotypes in the A#26-2; det3-1 mutant. In addition to the stunted plant stature mimicking det3-1, the A#26-2; det3-1 mutant exhibited stem thickening, increased floral organ number and a fruit shape reminiscent of clavata (clv) mutants. DNA sequencing analysis demonstrated that A#26-2; det3-1 harbors a mutation in the CLV3 gene. Importantly, A#26-2; det3-1 displayed cracks that randomly occurred on the main stem with a frequency of approximately 50%. Furthermore, the double mutants clv3-8 det3-1, clv1-4 det3-1 and clv2-1 det3-1 consistently showed stem cracks with frequencies of approximately 97, 38 and 35%, respectively. Cross-sections of stems further revealed an increase in vascular bundle number, cell number and size in the pith of clv3-8 det3-1 compared with det3-1. These findings suggest that the stem inner volume increase due to clv mutations exerts an outward mechanical stress; that in a det3-1 background (defective in cell expansion) resulted in cracking of the outermost layer of epidermal cells.

Pollen tubes of Cyrtanthus mackenii, a species with bicellular pollen, were cultured in vitro to investigate nuclear phase changes during generative cell division and male germ unit (MGU) formation, using flow cytometric analysis. Results revealed that sperm cells were formed after 12 h of culture. During sperm maturation, the nuclei of sperm cells were not associated with the vegetative nucleus (unassociated sperm cells; Sua) and became longer than those of sperm cells associated with the vegetative nucleus (Svn). These findings indicate that the pair of sperm cells in the C. mackenii MGU is dimorphic in terms of nuclear shape. Dimorphism coincides with anti-α-tubulin antibody immunofluorescence, which was higher in the Sua than in Svn. Following treatment with oryzalin, triggering microtubule depolymerization, differences between nuclear shapes in the two sperm nuclei disappeared, suggesting that microtubule accumulation between sperm cells in the MGU correlates with differences in the nuclear shape. © Springer-Verlag 2009.

This study aimed to analyze male gamete behavior from mature pollen to pollen tube growth in the bicellular pollen species Alstroemeria aurea. For mature pollen, pollen protoplasts were examined using flow cytometry. The protoplasts showed two peaks of DNA content at 1C and 1.90C. Flow cytometry at different developmental stages of pollen tubes cultured in vitro revealed changes in the nuclear phase at 9 and 18 h after culture. Sperm cell formation occurred at 6-9 h after culture, indicating that the first change was due to the division of the generative cells into sperm cells. After sperm cell formation, the number of vegetative nucleus associations with sperm cells showed a tendency to increase. This association was suggested as the male germ unit (MGU). When sperm cells, vegetative nuclei, and partial MGUs were collected separately from pollen tubes cultured for 18 h and analyzed using a flow cytometer, the sperm cells and vegetative nuclei contained 1C DNA, while the DNA content of partial MGUs was counted as 2C. Therefore, the second change in the nuclear phase, which results in an increase in 2C nuclei, is possibly related to the formation of MGUs. © 2009 The Botanical Society of Japan and Springer.
