Since the Fukushima Dai-ichi Nuclear Power Plant accident, radiation effects on nonhuman biota in the contaminated areas have been a major concern. Here, we analyzed the frequencies of chromosomal aberrations (translocations and dicentrics) in the splenic lymphocytes of large Japanese field mice (Apodemus speciosus) inhabiting Fukushima Prefecture. A. speciosus chromosomes 1, 2, and 5 were flow-sorted in order to develop A. speciosus chromosome-specific painting probes, and FISH (fluorescence in situ hybridization) was performed using these painting probes to detect the translocations and dicentrics. The average frequency of the translocations and dicentrics per cell in the heavily contaminated area was significantly higher than the frequencies in the case of the noncontaminated control area and the slightly and moderately contaminated areas, and this aberration frequency in individual mice tended to roughly increase with the estimated dose rates and accumulated doses. In all four sampling areas, the proportion of aberrations occurring in chromosome 2 was approximately > 3 times higher than that in chromosomes 1 and 5, which suggests that A. speciosus chromosome 2 harbors a fragile site that is highly sensitive to chromosome breaks induced by cellular stress such as DNA replication. The elevated frequency of chromosomal aberrations in A. speciosus potentially resulting from the presence of a fragile site in chromosome 2 might make it challenging to observe the mild effect of chronic low-dose-rate irradiation on the induction of chromosomal aberrations in A. speciosus inhabiting the contaminated areas of Fukushima.

© The Mammal Society of Japan. In some small mammals, digestive tract morphology is known to vary between and within individuals over time. Although changes in organs such as the stomach, small intestine, cecum, and large intestine vary among species, there is a trend toward increasing length and mass in individuals faced with increasing energy demands due to cold exposure or to reduced food quality. These morphological changes can also be induced by short day photoperiod, and could therefore serve as a winter adaptation. However, the extent to which these morphological responses to environmental cues are shared among mammals is not known. We examined the influence of cold temperature and short day photoperiod on the digestive tract morphology of Apodemus speciosus. The length and dry mass of all organs increased in response to cold temperature, and the dry mass of the small intestine was also increased by short day photoperiod. The small intestine is the principal site of nutrient absorption, and changes in its morphology may be an important aspect of winter adaptation. These results suggest that digestive tract morphology may alter flexibly as a means of coping with energy challenges.

© 2014. The mammalian gastrointestinal tract has evolved to facilitate food utilisation. Hamsters within the subfamily Cricetinae (Rodentia, Cricetidae) have a compartmentalised stomach consisting of a forestomach and a glandular stomach. The role of the forestomach in hamsters has long been discussed. In this study, we evaluated the microflora and volatile fatty acid contents of the large forestomach and caecum of a greater long-tailed hamster (Tscherskia triton). The estimated bacterial biodiversity in the forestomach based on 16S rRNA library sequencing analyses was low (sequence n=226, Shannon index H'=2.12) compared to that in the rumen of ruminants. In contrast, the bacterial diversity in the caecum was very high (n=259, H'=4.45), and comparable to that of other hindgut fermenters. The forestomach bacterial flora was dominated by Lactobacillus spp. (179/226 clones), and high concentrations of lactic acid were observed in the forestomach. These results indicate that the forestomach does not function like the rumen of ruminants, but that lactic acid fermentation does take place in this compartment. The caecum is thought to play a more important role in food digestion via fermentation by symbiotic microbes than the forestomach. When all cloned 16S rRNA gene sequences n(=485) were grouped into operational taxonomic units (OTUs) with 97% similarity, the majority of clones isolated from the forestomach contents were also present in the caecum (85.4%), although fewer clones isolated from the caecum contents shared OTUs with clones from the forestomach (19.3%). Based on these results, we hypothesise that bacteria were transferred from the caecum to the forestomach by coprophagy.
