ヒト腸管上皮細胞モデル (Caco-2) におけるキンカン由来β-クリプトキサンチンの膜透過性に及ぼす麹菌発酵乳飲料の影響

キンカンを加工し機能性成分であるβ-クリプトキサンチンの含有量を比較したところ, ペーストにより多くのβ-クリプトキサンチンが含まれ, ペーストと麹菌発酵乳飲料を混合しても, β-クリプトキサンチン含有量は変化しなかった. また, キンカンペースト添加麹菌発酵乳飲料を65 °C 及び 85 °Cで殺菌処理したところ, 65 °C試験区において多くのβ-クリプトキサンチンが残存することが示された. キンカン由来β-クリプトキサンチン-麹菌発酵乳飲料混合物のβ-クリプトキサンチン腸管吸収能を評価した結果, β-クリプトキサンチン単独区と比較し, 麹菌発酵乳飲料混合区の腸管吸収量は約14倍に上昇することが明らかとなった.

The chemical composition of muscles from Japanese eels cultured in hot-spring water was determined and compared with that of eels cultured in well water. The proximate composition did not differ significantly between the culture conditions, but the hepatosomatic indices of the hot-spring water eels were significantly higher than those of the well water eels. Amino acid levels were higher in the hot-spring water eels, and the carnosine content of the hot-spring eels was lower than that of the controls. Our results suggest that hot-spring water is a promising medium for eel culture, and its use may bring energy cost savings.

Inflammatory activity and hypoxia in atherosclerotic plaques are associated with plaque instability and thrombotic complications. Recent studies show that vascular cell metabolism affects atherogenesis and thrombogenicity. This study aimed to identify the metabolites in macrophage-rich unstable plaques that modulate atherogenesis and serve as potential markers of plaque instability. Atherosclerotic plaques were induced by balloon injury in the iliofemoral arteries of rabbits fed on a conventional or 0.5% cholesterol diet. At 3 months post-balloon injury, the arteries and cardiac tissues were subjected to histological, quantitative real-time polymerase chain reaction, and metabolomic analyses. The identified metabolite-related proteins were immunohistochemically analyzed in stable and unstable plaques from human coronary arteries. The factors modulating the identified metabolites were examined in macrophages derived from human peripheral blood mononuclear cells. Metabolomic analysis revealed that choline and guanine levels in macrophage-rich arteries were upregulated compared with those in non-injured arteries and cardiac tissues. Vascular choline levels, but not guanine levels, were positively correlated with the areas immunopositive for macrophages and tumor necrosis factor (TNF)-α and matrix metalloproteinase (MMP) 9 mRNA levels in injured arteries. In human coronary arteries, choline transporter-like protein (CTL) 1 was mainly localized to macrophages within plaques. The area that was immunopositive for CTL1 in unstable plaques was significantly higher than that in stable plaques. Intracellular choline levels were upregulated upon stimulation with TNF-α but were downregulated under hypoxia in cultured macrophages. Administration of choline upregulated the expression of TNF-α and CTL1 mRNA in cultured macrophages. The transfection of CTL1 small interfering RNA decreased CTL1, TNF-α, and MMP9 mRNA levels in cultured macrophages. These results suggest that choline metabolism is altered in macrophage-rich atherosclerotic lesions and unstable plaques. Thus, CTL1 may be potential markers of plaque instability.

Here we show a novel colorimetric sensor system with a photo-induced colorimetric reaction of N,N'-bis[2-(trimethylammonium)ethyl]-1,8:4,5-naphthalenetetracarboxdiimide (TENDI) to detect and quantify carboxylate derivatives. In the presence of CH3COONa, TENDI aqueous solution under UV irradiation shows a photo-induced colorimetric reaction, which is due to the production of naphthalenediimide (NDI)-based radical anions whose absorbance peaks appear at around 400–600 nm. In contrast, no color changes are observed in the cases of diverse inorganic salts. In addition, TENDI shows another ability to discriminate the structural features of carboxylate derivatives by color tone. Moreover, interestingly, the Hill equation can approximate the relationship between carboxylate concentrations and absorbance values on the maximum absorbance wavelengths of the absorbance peaks due to the NDI-radical anions.

Two new angular-type pyranocoumarins, namely hyuganin G (1) and hyuganin H (2), were isolated from an ethyl acetate extract of the aerial parts of Angelica furcijuga, together with three known angular-type pyranocoumarins, isoepoxypteryxin (3), isopteryxin (4), and hyuganin E (5). Assignment of the 1H and 13C NMR data of 1 and 2 were achieved via analysis of the DEPT, HMQC, and HMBC spectra. The absolute configurations of 1 and 2 were determined by semi-synthesis from 3.

© 2017 Elsevier Ltd Photocatalytic generation of H2 from water over titanium dioxide (TiO2) has received a great deal of interest for developing a renewable and clean energy source. It is initiated by charge-separation in TiO2 upon photoexcitation. The electron reduces water to generate H2 while the hole oxidizes hydroxide to generate hydroxyl radicals. However, water-splitting into O2 and H2 is not easy because of the large up-hill reaction and rapid reverse reaction. It is well known that the use of electron-donating sacrificial agents (hole scavengers) remarkably accelerates TiO2-photocatalyzed H2 evolution in which the hydroxyl radical is consumed by the sacrificial agents. Thus, sacrificial H2 production over a TiO2 photocatalyst is a convenient method to generate H2 from biomass and waste. This paper reviews the sacrificial H2 production from polyols, carboxylic acids, and saccharides, focusing on the chemical yield of H2 production.

A cobalt-free humidity indicator (HDI) for silica gel was developed using tetraphenylporphyrin (Tpp), which showed a distinguishable color change depending on pH. The HDI was prepared by mixing dichloro (tetraphenylporphyrinato)- phosphorus chloride (PTpp) and MgCl2 with SiO2 and drying at 130°C for 24 h. During the preparation, the PTpp was decomposed into the protonated Tpp (H2Tpp2+). The pH change arose from proton release by the reaction of MgCl2 with silanol of the SiO2 under dry conditions and neutralization under humid conditions. The HDI showed green color due to H2Tpp2+ under dry conditions and orange color due to Tpp under humid conditions. However, the HDI underwent partial decoloration on extended exposure to sunlight. Here, to prevent this decoloration, UV-absorbents coumarin and dibenzosuberenone were loaded onto the HDI. The HDIs with coumarin and without coumarin were irradiated at 352 nm and their microscopic absorption spectra were measured on a confocal laser scanning microscope. The absorbance at 650 nm and 515 nm of the HDI were monitored under dry and wet conditions, respectively. The light-protecting ability of UV-absorbent was analyzed by assuming that the decomposition process obeyed first-order kinetics. It was concluded that the additions of 0.125 wt% of coumarin and 0.050 wt% of dibenzosuberenone were effective to prevent decoloration of green color of H2Tpp2+ as well as orange color of Tpp.

© 2015 The Japan Society for Analytical Chemistry. The mass spectra of polycationic phosphorus porphyrin complexes were analyzed. It has been difficult to measure the mass spectra of polycationic phosphorus porphyrin complexes so far. Here, the mass spectra of a penta-cationic N,N′-dimethylbis[4-bipyridiniyl-3-propoxy]tetraphenylporphyrinato- phosphorus chloride bromide iodide complex (1a<sup>5+</sup>2Br<sup>−</sup>Cl<sup>−</sup>2I<sup>−</sup>) was successfully measured with a Fourier-transform mass spectrometer using electrospray ionization. A series of + 5, + 4, and + 3 complexes, generated by sequence reduction, were measured, and the isotope patterns of the complexes were matched with the calculated patterns. In the case of 1a<sup>5+</sup>5Cl<sup>−</sup>, which was treated by anion exchange, it was found that + 4 and + 3 were generated by deprotonation, but not sequence reduction. Therefore, sequence reduction was due to the presence of iodide ions. Polycationic porphyrin complexes are expected to be applicable to photodynamic therapy. The present measurements will contribute to the biological applications of polycationic compounds.

Bio-fuelization of herbaceous lignocelluloses through a simultaneous saccharification and fermentation process (SSF) and photocatalytic reforming (photo-Reform) was examined. The SSF of the alkali-pretreated bamboo, rice straw, and silvergrass was performed in an acetate buffer (pH 5.0) using cellulase, xylanase, and Saccharomyces cerevisiae at 34 °C. Ethanol was produced in 63%-85% yields, while xylose was produced in 74%-97% yields without being fermented because xylose cannot be fermented by S. cerevisiae. After the removal of ethanol from the aqueous SSF solution, the SSF solution was subjected to a photo-Reform step where xylose was transformed into hydrogen by a photocatalytic reaction using Pt-loaded TiO<inf>2</inf> (2 wt % of Pt content) under irradiation by a high pressure mercury lamp. The photo-Reform process produced hydrogen in nearly a yield of ten theoretical equivalents to xylose. Total energy was recovered as ethanol and hydrogen whose combustion energy was 73.4%-91.1% of that of the alkali-pretreated lignocelluloses (holocellulose). © 2014 by the authors.

Photocatalytic H2 evolution was examined using Pt-loaded TiO2-photocatalyst in the presence of amines as sacrificial agents. In the case of amines with all of the carbon attached to the hetero-atom such as 2-aminoethanol, 1,2-diamonoethane, 2-amino-1,3-propanediol, and 3-amino-1,2-propanediol, they were completely decomposed into CO2 and water to quantitatively evolve H2. On the other hand, the amines with both hetero-atoms and one methyl group at the β-positions (neighboring carbons) of amino group such as 2-amino-1-propanol and 1,2-diaminopropane were partially decomposed. Also, the photocatalytic H2 evolution using amines without the hetero-atoms at the β-positions such as ethylamine, propylamine, 1-butylamine, 1,3-diaminopropane, 2-propylamine, and 2-butylamine was inefficient. Thus, it was found that the neighboring hetero-atom strongly assisted the degradation of sacrificial amines. Moreover, rate constants for H2 evolution were compared among amines. In conclusion, the neighboring hetero-atom did not affect the rate constants but enhanced the yield of hydrogen evolution. © 2014 by the authors; licensee MDPI, Basel, Switzerland.

The photocatalytic H2 evolution by Pt-loaded TiO2 was examined using sacrificial agents such as 1,2-ethanediol, glycerol, erythritol, and arabitol, the carbons of which all have hydroxy groups. In these cases, almost complete decomposition of sacrificial agents into CO2 and H 2O occurred. Moreover, the H2 evolution reached the maximum values estimated by their potentially electron-donating ability. On the other hand, smaller amounts of CO2 and H2 were evolved from 1,2,3-butanetriol, 1,2,4-butanetriol, and 1,2-butanediol, which had both continuous hydroxylated carbons and one or two non-hydroxy-substituted carbons, the inner continuous 2,3-butanediol, and the discontinuous 1,3- and 1,4- butanediols. In the cases of monools such as 2-butanol and 2-propanol, CO2 was not evolved at all. Thus, we found that the structure of sacrificial agents, more so than the potential electron-donating ability, is an important factor to evolve H2 efficiently. © 2013 The Chemical Society of Japan.

The acidity of magnesium salts-silica gel composites was measured by absorption spectrophotometry in the micro-region using a confocal laser scanning microscope (CLSM). It has been reported that MgCl2-SiO2releases protons under dry conditions, and goes back to neutral under wet conditions. At first, using five kinds of tetraarylporphyrins, the absorbance (AP) at 515 nm due to a free base type and that (AG) at 650 nm due to the protonated porphyrins were measured in a CHCl3-MeOH-water solution, whose apparent pH was adjusted by HClO4. Fraction (F-value) of AP [F = AP/(AG+ AP)] was calculated at every apparent pH to produce apparent pH-profiles of the f-values for all porphyrins. The profiles were fitted by sigmoid curves. Second, the porphyrin was adsorbed on MgCl2-SiO2. The porphyrin- adsorbed composite was kept under the given relative humidity conditions for 3 days and subjected to CLSM analysis. The F-values of the porphyrins adsorbed on the composites were obtained from the absorption spectra to determine the apparent pH using fitting curves. As a result, the MgCl2-SiO2had an apparent pH of 1.73 under dry conditions, and 4.39 under wet conditions. Moreover, the pH values of the Mg(NO3)2-SiO2composite and the MgSO4-SiO2composite were measured. © 2012, The Japan Society for Analytical Chemistry. All rights reserved.

A sensitive and easily distinguishable cobalt-free humidity indicator of porphyrin-silica gel-MgCl(2) composite was prepared from pH-induced spectra changeable tetraarylporphyrin, silica gel (SiO(2)), and MgCl(2). The pH change arose from proton release under dry conditions, and proton capture under humid conditions by a reversible reaction between MgCl(2) and a silanol group of SiO(2). A pink-orange porphyrin-Si(OH)(2)-MgCl(2) composite was dried to give a green protonated porphyrin-SiO(2)Mg composite. The optimized concentrations of MgCl(2) to make the concentrations of protonated porphyrin maximum under dry conditions were determined by absorption spectrometry of the green composite using a confocal laser scanning microscope as a microscopic spectrometer. Moreover, the green composite was prepared by heating dichloro(tetraarylporphyrinato)phosphorus chloride with MgCl(2) and SiO(2). The humidity-sensitivity of the green composite was evaluated by the absorption spectra under controlled humidity. A distinguishable color change of the green composite took place below 30% of relative humidity.

In an effort to develop water-soluble singlet-oxygen (1O 2) photosensitizer, we synthesized a series of phosphorus porphyrin complexes (1, (RO)2P(tpp)+Cl-, tpp = tetraphenylporphyrinato ligand, R = -(CH2CH2O) m-(CH2)n-H, m = 0-3, n = 1, 2, 4, 6) and evaluated their performance characteristics. Water-solubilities of 1 (m = 1, 2 and n = 1, 2, 4, 6) were determined to be >1.0 mM, which was higher than the parent (HO)2P(tpp)+Cl-. The quantum yields for the generation of 1O2 in water based on the amount of 11* were determined to be 0.73, 0.69, and 0.62 for 1d (m = 2, n = 4), 1e (m = 2, n = 2), and 1f (m = 2, n = 1), respectively. Photo-inactivation by 1 was performed with yeast (Saccharomyces cerevisiae), and was evaluated using a inactivation factor AF = ([1]M × T1/2)-1, where [1]M is the minimum effective concentration of 1 and T1/2 is a half-life of the yeast initially used. In the case of 1b (m = 1, n = 6) and 1c (m = 2, n = 6), [1] M was determined to be 5 nM in the photo-sterilization. Bio-affinity was evaluated by the adsorbed concentration of 1 ([P]ad) into the yeast, which was measured by absorption spectrophotometry using a confocal laser-scanning microscope. These findings revealed that a large amount of 1 was adsorbed into the yeast. The estimated amount of 1b obtained from 10 μM aqueous solution came to 171 mM (17,100 fold) in terms of concentration. As an increase in carbon number (n) of the alkyl group on axial ligands, AF and [P]ad increased. It is strongly suggested that the photo-inactivation took place with the assistance of the attack of 1O2 inside the cells. © 2011 Elsevier B.V.

An evaluation of the bio-affinity of methoxo(alkyloxo)-tetraphenylporphyrinatoantimony bromide (1) was examined by an absorption experiment of 1 toward Saccharomyces cerevisiae. An aqueous solution of 1 was added to an aqueous solution of S. cerevisiae. Then an aqueous solution of agar was added into the solution in order to stop the Brownian motion of bacteria during analysis. A prepared aqueous solution containing 1(10 μmol dm-3), S. cerevisiae (1.0 ×104 cells cm-3) and agar (0.4 wt%) was taken inside a space surrounded by a silicone spacer (1 cm ×1 cm, thickness 50 μm) put on a slide glass, which was set on a confocal laser scanning microscope (CLSM) stage. By measuring the diameter of the cell and the absorbance on CLSM, the concentrations of 1 adsorbed inside S. cerevisia were determined to be 54.2 ±7.3 for 1a, 49.8 ±6.2 for 1b, and 22.5 ±15.5 mmol dm-3 for 1c according to Lambert-Beer's law. © 2009 The Japan Society for Analytical Chemistry.

Sterilization activities of seven kinds of water-soluble dialkyloxo(tetraphenylporphyrinato)antimony(V) bromides (1) were examined against a yeast, Saccharomyces cerevisiae, under visible light irradiation. Sterilization activities were analyzed by the first-order kinetics involving the generation of singlet oxygen (1O2) in the complex between 1 and the cells. The formation of the complexes was confirmed by the measurements of the saturated adsorption concentrations of 1 inside yeast by absorption spectrophotometry using a confocal laser scanning microscope. The quantum yields of 1 for the generation of 1O2 were determined to be 0.48-0.65. In order to ensure efficient sterilization, the requisite was that 1 were both water-soluble and oil-soluble. Presumably the oil-soluble feature of 1 was advantageous in passing through the cell wall of the yeast which consisted of hydrophobic peptideglucan and the water-soluble feature was advantageous in the adsorption occurring at hydrophilic sites inside the cell, causing fatal damage to the yeast. © 2009 Elsevier B.V. In order to elucidate the applicable range and accuracy of absorption spectrophotometry in the micro-region using a confocal laser scanning microscope (CLSM), micro-silica gel beads (2) immobilizing a given molar concentration (C1m) of dihydroxo(tetraphenylporphyrinato)-antimony(V) bromide (1) were prepared. The measurable area on the beads was determined to be a circle of 2.14 μm diameter by magnifications of an objective lens, the diameter of fiber core, and the diameter of a pinhole. Moreover, the range of the diameter of the beads and the spectral range were decided to be μm ∼ 350 μm and 488-775 nm, respectively. Using a halogen lamp as a light source on CLSM, the molar concentrations (C2) of 1 in 2 were determined by absorption spectrophotometry. The value of C2 showed good agreement with C1m with 0.11 -0.73 of the standard deviation. Thus, absorption spectrophotometry in the micro-region will be a powerful tool for the quantitative analysis of transparent materials of micron-order. © 2008 The Japan Society for Analytical Chemistry.

Micro-silica gel beads (1a-c) immobilizing dihydroxoantimony(V) tetraphenylporphyrin (SbTpp), free base tetraphenylporphyrin (H2Tpp), and zinc(II) tetraphenylporphyrin (ZnTpp) chromophores were prepared by the reactions of 3-aminopropyl silica gel (SiO2-NH2) with the dihydroxoantimony(V) complex (2a), the free base (2b), and the zinc complex (2c) of 5-[4-(succinimidyloxycarbonyl)phenyl]-10,15,20-triphenylporphyrin, respectively. Absorption spectrophotometry was performed on 1a-c using a confocal laser scanning microscope (CLSM) and determined that the yields for the immobilization of SbTpp, H2Tpp, and ZnTpp on SiO2-NH2 were 31.7, 95.9, and 45.1%, respectively. Moreover, 1a was converted into its acetyl analog (1a') by the acetylation of the axial hydroxo ligands and aminopropyl of 1a with Ac2O, since the interactions of the SbTpp chromophore with the residual amino group on SiO2 was observed by spectroscopic analysis using CLSM. In micro-channel reactors (MCR), the photoreaction of 1a' with Et2NH induced demetallation to produce 1b immobilizing H2Tpp chromophore. Moreover, the reaction of 1b with Zn(OAc)2 produced 1c immobilizing ZnTpp chromophore. Thus, micro-silica gel beads (1a) were transformed into 1c in a micro-region of a silica gel surface by way of photochemical reaction with Et2NH and subsequent metallation with Zn(OAc)2. © 2008 Elsevier B.V. Dihydroxoantimony(V) tetraphenylporphyrin chromophore (SbTPP) was immobilized on 3-aminopropyl-silica gel (NH 2-SiO 2) by the reaction of dihydroxoantimony [4-(N-succinimidyloxycarbonyl)phenyl] triphenylporphyrin with NH 2-SiO 2. Spectroscopic analysis of the complex by confocal laser scanning microscope suggested the interaction of SbTPP with the residual amino group on SiO 2.
