ヒト腸管上皮細胞モデル (Caco-2) におけるキンカン由来β-クリプトキサンチンの膜透過性に及ぼす麹菌発酵乳飲料の影響

キンカンを加工し機能性成分であるβ-クリプトキサンチンの含有量を比較したところ, ペーストにより多くのβ-クリプトキサンチンが含まれ, ペーストと麹菌発酵乳飲料を混合しても, β-クリプトキサンチン含有量は変化しなかった. また, キンカンペースト添加麹菌発酵乳飲料を65 °C 及び 85 °Cで殺菌処理したところ, 65 °C試験区において多くのβ-クリプトキサンチンが残存することが示された. キンカン由来β-クリプトキサンチン-麹菌発酵乳飲料混合物のβ-クリプトキサンチン腸管吸収能を評価した結果, β-クリプトキサンチン単独区と比較し, 麹菌発酵乳飲料混合区の腸管吸収量は約14倍に上昇することが明らかとなった.

The chemical composition of muscles from Japanese eels cultured in hot-spring water was determined and compared with that of eels cultured in well water. The proximate composition did not differ significantly between the culture conditions, but the hepatosomatic indices of the hot-spring water eels were significantly higher than those of the well water eels. Amino acid levels were higher in the hot-spring water eels, and the carnosine content of the hot-spring eels was lower than that of the controls. Our results suggest that hot-spring water is a promising medium for eel culture, and its use may bring energy cost savings.

Inflammatory activity and hypoxia in atherosclerotic plaques are associated with plaque instability and thrombotic complications. Recent studies show that vascular cell metabolism affects atherogenesis and thrombogenicity. This study aimed to identify the metabolites in macrophage-rich unstable plaques that modulate atherogenesis and serve as potential markers of plaque instability. Atherosclerotic plaques were induced by balloon injury in the iliofemoral arteries of rabbits fed on a conventional or 0.5% cholesterol diet. At 3 months post-balloon injury, the arteries and cardiac tissues were subjected to histological, quantitative real-time polymerase chain reaction, and metabolomic analyses. The identified metabolite-related proteins were immunohistochemically analyzed in stable and unstable plaques from human coronary arteries. The factors modulating the identified metabolites were examined in macrophages derived from human peripheral blood mononuclear cells. Metabolomic analysis revealed that choline and guanine levels in macrophage-rich arteries were upregulated compared with those in non-injured arteries and cardiac tissues. Vascular choline levels, but not guanine levels, were positively correlated with the areas immunopositive for macrophages and tumor necrosis factor (TNF)-α and matrix metalloproteinase (MMP) 9 mRNA levels in injured arteries. In human coronary arteries, choline transporter-like protein (CTL) 1 was mainly localized to macrophages within plaques. The area that was immunopositive for CTL1 in unstable plaques was significantly higher than that in stable plaques. Intracellular choline levels were upregulated upon stimulation with TNF-α but were downregulated under hypoxia in cultured macrophages. Administration of choline upregulated the expression of TNF-α and CTL1 mRNA in cultured macrophages. The transfection of CTL1 small interfering RNA decreased CTL1, TNF-α, and MMP9 mRNA levels in cultured macrophages. These results suggest that choline metabolism is altered in macrophage-rich atherosclerotic lesions and unstable plaques. Thus, CTL1 may be potential markers of plaque instability.

Here we show a novel colorimetric sensor system with a photo-induced colorimetric reaction of N,N'-bis[2-(trimethylammonium)ethyl]-1,8:4,5-naphthalenetetracarboxdiimide (TENDI) to detect and quantify carboxylate derivatives. In the presence of CH3COONa, TENDI aqueous solution under UV irradiation shows a photo-induced colorimetric reaction, which is due to the production of naphthalenediimide (NDI)-based radical anions whose absorbance peaks appear at around 400–600 nm. In contrast, no color changes are observed in the cases of diverse inorganic salts. In addition, TENDI shows another ability to discriminate the structural features of carboxylate derivatives by color tone. Moreover, interestingly, the Hill equation can approximate the relationship between carboxylate concentrations and absorbance values on the maximum absorbance wavelengths of the absorbance peaks due to the NDI-radical anions.

Two new angular-type pyranocoumarins, namely hyuganin G (1) and hyuganin H (2), were isolated from an ethyl acetate extract of the aerial parts of Angelica furcijuga, together with three known angular-type pyranocoumarins, isoepoxypteryxin (3), isopteryxin (4), and hyuganin E (5). Assignment of the 1H and 13C NMR data of 1 and 2 were achieved via analysis of the DEPT, HMQC, and HMBC spectra. The absolute configurations of 1 and 2 were determined by semi-synthesis from 3.

© 2017 Elsevier Ltd Photocatalytic generation of H2 from water over titanium dioxide (TiO2) has received a great deal of interest for developing a renewable and clean energy source. It is initiated by charge-separation in TiO2 upon photoexcitation. The electron reduces water to generate H2 while the hole oxidizes hydroxide to generate hydroxyl radicals. However, water-splitting into O2 and H2 is not easy because of the large up-hill reaction and rapid reverse reaction. It is well known that the use of electron-donating sacrificial agents (hole scavengers) remarkably accelerates TiO2-photocatalyzed H2 evolution in which the hydroxyl radical is consumed by the sacrificial agents. Thus, sacrificial H2 production over a TiO2 photocatalyst is a convenient method to generate H2 from biomass and waste. This paper reviews the sacrificial H2 production from polyols, carboxylic acids, and saccharides, focusing on the chemical yield of H2 production.

A cobalt-free humidity indicator (HDI) for silica gel was developed using tetraphenylporphyrin (Tpp), which showed a distinguishable color change depending on pH. The HDI was prepared by mixing dichloro (tetraphenylporphyrinato)- phosphorus chloride (PTpp) and MgCl2 with SiO2 and drying at 130°C for 24 h. During the preparation, the PTpp was decomposed into the protonated Tpp (H2Tpp2+). The pH change arose from proton release by the reaction of MgCl2 with silanol of the SiO2 under dry conditions and neutralization under humid conditions. The HDI showed green color due to H2Tpp2+ under dry conditions and orange color due to Tpp under humid conditions. However, the HDI underwent partial decoloration on extended exposure to sunlight. Here, to prevent this decoloration, UV-absorbents coumarin and dibenzosuberenone were loaded onto the HDI. The HDIs with coumarin and without coumarin were irradiated at 352 nm and their microscopic absorption spectra were measured on a confocal laser scanning microscope. The absorbance at 650 nm and 515 nm of the HDI were monitored under dry and wet conditions, respectively. The light-protecting ability of UV-absorbent was analyzed by assuming that the decomposition process obeyed first-order kinetics. It was concluded that the additions of 0.125 wt% of coumarin and 0.050 wt% of dibenzosuberenone were effective to prevent decoloration of green color of H2Tpp2+ as well as orange color of Tpp.

© 2015 The Japan Society for Analytical Chemistry. The mass spectra of polycationic phosphorus porphyrin complexes were analyzed. It has been difficult to measure the mass spectra of polycationic phosphorus porphyrin complexes so far. Here, the mass spectra of a penta-cationic N,N′-dimethylbis[4-bipyridiniyl-3-propoxy]tetraphenylporphyrinato- phosphorus chloride bromide iodide complex (1a<sup>5+</sup>2Br<sup>−</sup>Cl<sup>−</sup>2I<sup>−</sup>) was successfully measured with a Fourier-transform mass spectrometer using electrospray ionization. A series of + 5, + 4, and + 3 complexes, generated by sequence reduction, were measured, and the isotope patterns of the complexes were matched with the calculated patterns. In the case of 1a<sup>5+</sup>5Cl<sup>−</sup>, which was treated by anion exchange, it was found that + 4 and + 3 were generated by deprotonation, but not sequence reduction. Therefore, sequence reduction was due to the presence of iodide ions. Polycationic porphyrin complexes are expected to be applicable to photodynamic therapy. The present measurements will contribute to the biological applications of polycationic compounds.
