The acidity of magnesium salts-silica gel composites was measured by absorption spectrophotometry in the micro-region using a confocal laser scanning microscope (CLSM). It has been reported that MgCl2-SiO2releases protons under dry conditions, and goes back to neutral under wet conditions. At first, using five kinds of tetraarylporphyrins, the absorbance (AP) at 515 nm due to a free base type and that (AG) at 650 nm due to the protonated porphyrins were measured in a CHCl3-MeOH-water solution, whose apparent pH was adjusted by HClO4. Fraction (F-value) of AP [F = AP/(AG+ AP)] was calculated at every apparent pH to produce apparent pH-profiles of the f-values for all porphyrins. The profiles were fitted by sigmoid curves. Second, the porphyrin was adsorbed on MgCl2-SiO2. The porphyrin- adsorbed composite was kept under the given relative humidity conditions for 3 days and subjected to CLSM analysis. The F-values of the porphyrins adsorbed on the composites were obtained from the absorption spectra to determine the apparent pH using fitting curves. As a result, the MgCl2-SiO2had an apparent pH of 1.73 under dry conditions, and 4.39 under wet conditions. Moreover, the pH values of the Mg(NO3)2-SiO2composite and the MgSO4-SiO2composite were measured. © 2012, The Japan Society for Analytical Chemistry. All rights reserved.

A sensitive and easily distinguishable cobalt-free humidity indicator of porphyrin-silica gel-MgCl(2) composite was prepared from pH-induced spectra changeable tetraarylporphyrin, silica gel (SiO(2)), and MgCl(2). The pH change arose from proton release under dry conditions, and proton capture under humid conditions by a reversible reaction between MgCl(2) and a silanol group of SiO(2). A pink-orange porphyrin-Si(OH)(2)-MgCl(2) composite was dried to give a green protonated porphyrin-SiO(2)Mg composite. The optimized concentrations of MgCl(2) to make the concentrations of protonated porphyrin maximum under dry conditions were determined by absorption spectrometry of the green composite using a confocal laser scanning microscope as a microscopic spectrometer. Moreover, the green composite was prepared by heating dichloro(tetraarylporphyrinato)phosphorus chloride with MgCl(2) and SiO(2). The humidity-sensitivity of the green composite was evaluated by the absorption spectra under controlled humidity. A distinguishable color change of the green composite took place below 30% of relative humidity.

In an effort to develop water-soluble singlet-oxygen (1O 2) photosensitizer, we synthesized a series of phosphorus porphyrin complexes (1, (RO)2P(tpp)+Cl-, tpp = tetraphenylporphyrinato ligand, R = -(CH2CH2O) m-(CH2)n-H, m = 0-3, n = 1, 2, 4, 6) and evaluated their performance characteristics. Water-solubilities of 1 (m = 1, 2 and n = 1, 2, 4, 6) were determined to be >1.0 mM, which was higher than the parent (HO)2P(tpp)+Cl-. The quantum yields for the generation of 1O2 in water based on the amount of 11* were determined to be 0.73, 0.69, and 0.62 for 1d (m = 2, n = 4), 1e (m = 2, n = 2), and 1f (m = 2, n = 1), respectively. Photo-inactivation by 1 was performed with yeast (Saccharomyces cerevisiae), and was evaluated using a inactivation factor AF = ([1]M × T1/2)-1, where [1]M is the minimum effective concentration of 1 and T1/2 is a half-life of the yeast initially used. In the case of 1b (m = 1, n = 6) and 1c (m = 2, n = 6), [1] M was determined to be 5 nM in the photo-sterilization. Bio-affinity was evaluated by the adsorbed concentration of 1 ([P]ad) into the yeast, which was measured by absorption spectrophotometry using a confocal laser-scanning microscope. These findings revealed that a large amount of 1 was adsorbed into the yeast. The estimated amount of 1b obtained from 10 μM aqueous solution came to 171 mM (17,100 fold) in terms of concentration. As an increase in carbon number (n) of the alkyl group on axial ligands, AF and [P]ad increased. It is strongly suggested that the photo-inactivation took place with the assistance of the attack of 1O2 inside the cells. © 2011 Elsevier B.V.

An evaluation of the bio-affinity of methoxo(alkyloxo)-tetraphenylporphyrinatoantimony bromide (1) was examined by an absorption experiment of 1 toward Saccharomyces cerevisiae. An aqueous solution of 1 was added to an aqueous solution of S. cerevisiae. Then an aqueous solution of agar was added into the solution in order to stop the Brownian motion of bacteria during analysis. A prepared aqueous solution containing 1(10 μmol dm-3), S. cerevisiae (1.0 ×104 cells cm-3) and agar (0.4 wt%) was taken inside a space surrounded by a silicone spacer (1 cm ×1 cm, thickness 50 μm) put on a slide glass, which was set on a confocal laser scanning microscope (CLSM) stage. By measuring the diameter of the cell and the absorbance on CLSM, the concentrations of 1 adsorbed inside S. cerevisia were determined to be 54.2 ±7.3 for 1a, 49.8 ±6.2 for 1b, and 22.5 ±15.5 mmol dm-3 for 1c according to Lambert-Beer's law. © 2009 The Japan Society for Analytical Chemistry.

Sterilization activities of seven kinds of water-soluble dialkyloxo(tetraphenylporphyrinato)antimony(V) bromides (1) were examined against a yeast, Saccharomyces cerevisiae, under visible light irradiation. Sterilization activities were analyzed by the first-order kinetics involving the generation of singlet oxygen (1O2) in the complex between 1 and the cells. The formation of the complexes was confirmed by the measurements of the saturated adsorption concentrations of 1 inside yeast by absorption spectrophotometry using a confocal laser scanning microscope. The quantum yields of 1 for the generation of 1O2 were determined to be 0.48-0.65. In order to ensure efficient sterilization, the requisite was that 1 were both water-soluble and oil-soluble. Presumably the oil-soluble feature of 1 was advantageous in passing through the cell wall of the yeast which consisted of hydrophobic peptideglucan and the water-soluble feature was advantageous in the adsorption occurring at hydrophilic sites inside the cell, causing fatal damage to the yeast. © 2009 Elsevier B.V. In order to elucidate the applicable range and accuracy of absorption spectrophotometry in the micro-region using a confocal laser scanning microscope (CLSM), micro-silica gel beads (2) immobilizing a given molar concentration (C1m) of dihydroxo(tetraphenylporphyrinato)-antimony(V) bromide (1) were prepared. The measurable area on the beads was determined to be a circle of 2.14 μm diameter by magnifications of an objective lens, the diameter of fiber core, and the diameter of a pinhole. Moreover, the range of the diameter of the beads and the spectral range were decided to be μm ∼ 350 μm and 488-775 nm, respectively. Using a halogen lamp as a light source on CLSM, the molar concentrations (C2) of 1 in 2 were determined by absorption spectrophotometry. The value of C2 showed good agreement with C1m with 0.11 -0.73 of the standard deviation. Thus, absorption spectrophotometry in the micro-region will be a powerful tool for the quantitative analysis of transparent materials of micron-order. © 2008 The Japan Society for Analytical Chemistry.

Micro-silica gel beads (1a-c) immobilizing dihydroxoantimony(V) tetraphenylporphyrin (SbTpp), free base tetraphenylporphyrin (H2Tpp), and zinc(II) tetraphenylporphyrin (ZnTpp) chromophores were prepared by the reactions of 3-aminopropyl silica gel (SiO2-NH2) with the dihydroxoantimony(V) complex (2a), the free base (2b), and the zinc complex (2c) of 5-[4-(succinimidyloxycarbonyl)phenyl]-10,15,20-triphenylporphyrin, respectively. Absorption spectrophotometry was performed on 1a-c using a confocal laser scanning microscope (CLSM) and determined that the yields for the immobilization of SbTpp, H2Tpp, and ZnTpp on SiO2-NH2 were 31.7, 95.9, and 45.1%, respectively. Moreover, 1a was converted into its acetyl analog (1a') by the acetylation of the axial hydroxo ligands and aminopropyl of 1a with Ac2O, since the interactions of the SbTpp chromophore with the residual amino group on SiO2 was observed by spectroscopic analysis using CLSM. In micro-channel reactors (MCR), the photoreaction of 1a' with Et2NH induced demetallation to produce 1b immobilizing H2Tpp chromophore. Moreover, the reaction of 1b with Zn(OAc)2 produced 1c immobilizing ZnTpp chromophore. Thus, micro-silica gel beads (1a) were transformed into 1c in a micro-region of a silica gel surface by way of photochemical reaction with Et2NH and subsequent metallation with Zn(OAc)2. © 2008 Elsevier B.V.
