Previous studies have shown the physiological significance of dietary conjugated linoleic acid (CLA) in various experimental animals and in human beings. One of the important problems to better elucidate is the difference between triglyceride (TG) and free (FFA) dietary CLA. Here, using splenocytes, this study assesses how TG- and FFA-CLA modulate immunoglobulin and various cytokine productions. In this study, C57BL/6N mice were fed an experimental diet containing 0% CLA, 0.1 or 1% FFA-CLA, or 0.1 or 1% TG-CLA for 3 weeks. The production of immunoglobulin tended to be up-regulated by 1% FFA-CLA. As a result of protein array analysis using the supernatant from splenocytes cultured with no CLA, 1% FFA-CLA, and TG-CLA, some cytokine production was shown to be remarkably regulated by dietary FFA- and TG-CLA. A total of 32 cytokines were examined, and 11-14 produced cytokines that were 2-fold up-regulated as compared with control for FFA- or TG-CLA, respectively. Especially, the production of IL-9 and MCP-5 and other cytokines was remarkably up-regulated by both FFA- and TG-CLA. In addition, seven cytokines were 2-fold down-regulated by TG-CLA. These data show that there is a slight but significant difference between the functionalities of FFA- and TG-CLA.

We evaluated the effect of conjugated linoleic acid (CLA) isomers on the growth factor-induced proliferation of human breast cancer MCF-7 cells. When MCF-7 cells were cultured in RPMI 1640 medium supplemented with 1% fetal bovine serum (FBS), CLA inhibited the proliferation and notably cis9, trans11 (c9,t11)-CLA showed the strongest effect. However, cells barely grew when cultured with 1% charcoal-treated FBS (cFBS). Proliferation was promoted in cFBS cultured cells by the addition of 17β-estradiol (E2), insulin, and epidermal growth factor (EGF). Trans10, cis12 (T10,c12)-CLA inhibited cell proliferation induced by E2 and insulin, but not by EGF. T10,c12-CLA also exhibited cell-killing activity when cells were induced with insulin. On the other hand, c9,t11-CLA was shown to have no effect on MCF-7 cell proliferation induced by and of these three growth factors. In conclusion, although both c9, t11 and t10, c12-CLA can inhibit the proliferation of MCF-7 cells, our results suggested that they have separate mechanisms and different targets of actions. © 2003 Elsevier Ireland Ltd. We showed the inhibitory effect of oleic and palmitoleic acids (OA and POA) on this trans10, cis12 (10t, 12c)-CLA induced cytotoxic activity. When cells were cultured in the presence of 10t, 12c-CLA, this potent cytotoxic effect on dRLh-84 cells was clearly obvious when compared to the control vehicle group. It was revealed that the levels of cellular OA and POA levels decreased upon cis9, trans11 (9c,11t) or 10t, 12c-CLA treatment in a time course dependent manner. OA or POA demonstrates a dose dependent inhibition of the 10t, 12c-CLA induced cytotoxicity. Notable nuclear fragmentation or activation of caspase-3 and 9 by 10t, 12c-CLA in dRLh-84 was counteracted by treatment with OA or POA. Results also suggest that 10t, 12c-CLA induced apoptosis can be inhibited by treatment with OA or POA. © 2003 Elsevier Science Ltd. All rights reserved.

We evaluated the effect of cis-9, trans-11 (9c, 11t) and trans-10, cis-12 (10t, 12c) conjugated linoleic acid (CLA) on the immune system in C57BL/6J mice. Mice were fed experimental diets containing 0% CLA (controls), 1% 9c, 11t-CLA, 1% 10t, 12c-CLA or a 1:1 mixture (0.5% + 0.5%) of these two CLA isomers for 3 wk. Relative spleen weights of all CLA fed mice were greater than the controls. Spleen lymphocytes isolated from the mice fed 10t, 12c-CLA produced more immunoglobulin (Ig)A and IgM but not IgG when stimulated with concanavalin A (ConA) compared with controls. IgA production from unstimulated spleen lymphocytes was greater in the 10t, 12c-CLA group than in controls. Conversely, 9c, 11t-CLA did not affect the production of any of the Ig subclasses. Lymphocytes isolated from 9c, 11t-CLA fed mice produced more tumor necrosis factor-α than the control group. The proportion of B cells in the spleen lymphocyte population was significantly lower in the 9c, 11t-CLA group, and higher in the 10t, 12c-CLA group than in the controls. Compared with the control group, the percentage of CD4 + T cells was lower in the 10t, 12c-CLA group, and the percentage of CD8 + T cells was higher in the 9c, 11t-CLA group. Furthermore, the percentage of CD8 + T cells was higher in the 1:1 mixture group than in controls. The CD4 + /CD8 + ratio was lower in the 1:1 mixture group than in controls. These results suggest that 9c, 11t and 10t, 12c-CLA can stimulate different immunological effects and that the simultaneous intake of the two isomers can change the T cell population.

OBJECTIVES: In this study, we examined the effect of dietary conjugated linoleic acid (CLA) on body fat levels in Sprague-Dawley rats. Methods: Rats were fed AIN-93G type diets containing 4%, 7%, and 10% fats with or without 1.5% CLA. Results: Three weeks after the onset of the experimental period, the weights of perirenal white adipose tissue were lower in CLA-fed rats. The weights of epididymal white adipose tissue also were lower in CLA-fed rats than in control rats, but this effect disappeared with increased dietary fat level. Serum leptin levels tended to be lower in the CLA group, especially the low-fat diet group, than in the control group. There were significant positive correlations between serum leptin level and weights of perirenal and epididymal white adipose tissues in control groups, but these correlations were weaker in the CLA groups. Serum tumor necrosis factor-α levels also tended to be lower in CLA-fed rats, and this tendency was most remarkable in the rats fed 7% fat diets. Conclusions: In conclusion, dietary CLA, especially the low-fat diet, reduced body fat without hepatic injury to Sprague-Dawley rats. © Elsevier Science Inc. 2003.

We evaluated the cytotoxic effect of conjugated linoleic acid (CLA) on rat hepatoma dRLh-84 cells in vitro. When cells were cultured in the presence of CLA, strong cytotoxic effect on dRLh-84 cells was recognized at 1μM level compared to the control vehicle group, and trans10, cis12-CLA but not cis9, trans11-CLA was shown to be an active isomer for inducing this effect. Increase of the sub-G1 population and activation of caspase-3 and 9 accompanied with a time-dependent cleavage of poly(ADP-ribose) polymerase were recognized in dRLh-84 cells treated with trans10, cis12-CLA. In addition, we could see nuclear fragmentation in dRLh-84 cells treated with trans10, cis12-CLA by laser scanning confocal microscopy observation. Cytotoxic effect of trans10, cis12-CLA on normal hepatocytes was weaker than on dRLh-84 cells. These data indicate trans10, cis12-CLA has a potent cytotoxic effect on dRLh-84 cells through at least in part by an apoptotic pathway. © 2002 Elsevier Science Ireland Ltd. All rights reserved.

We examined the effect of dietary conjugated linoleic acid (CLA) on the growth of injected hepatoma dRLh-84 in Donryu rats. After experimental diets containing 0% or 2% CLA were given to male Donryu rats for 3 wk, dRLh-84 cells were injected into the left lobe of the hepatic capsule, and the experimental diet was continued. The cells formed a solid tumor ≥1 wk after the injection, and thereafter the tumor grew with feeding duration. In a morphological study, this tumor appeared to be a low-differentiated hepatoma, and there was no remarkable difference in the morphology of the tumor between 0% and 2% CLA groups. Tumor weight was significantly higher in the 2% CLA group than in the 0% CLA group throughout the feeding period after the injection. Serum glutamic-oxaloacetic transaminase and glutamic-pyruvic transaminase activities were significantly higher in 2% CLA-injected rats than in 0% CLA-injected rats at 3 wk after the injection. CLA upregulated acyl-CoA oxidase activity, especially 1 wk after the injection. However, dietary CLA did not activate carnitine palmitoyl transferase II, which is a rate-limiting enzyme in the mitochondrial β-oxidation pathway. Natural killer cell activity in the spleen tended to be higher in injected rats, but a significant effect of dietary CLA was not recognized. Serum interferon-γ and tumor necrosis factor-α levels were higher in injected than in sham rats. More-over, these levels were higher in 2% CLA groups than in the respective 0% CLA groups.
